Applications of BBa_K5213001

This part was intended to be used to produce brominated substances that are able to inhibit methanogenesis. It was intended to be coupled with part BBa_K5213001 under the control of promoter BBa_K5213003 that would constitutively deliver the necessary substrate H2O2 that is present at low levels within prokaryotic cells due to toxicity. Additionally, it encodes a 6xHis tag at the N-terminal of the protein that should not interact with the conformation based on structural analysis however can be taken advantage of when purifying.

The amplification of the part with the designated primer sets did not occur properly, with unspecific by-product formation. Alterations of primer, template and annealing temperature did not prevent the formation of the unspecific product. Different PCR types such as touchdown and inverse PCR also did not yield acceptable results. Attempt at extracting the correct fragment from the gel yielded very low results, that could not be used within our plasmid assembly. Hence work with the part was terminated and it could not be characterized.
